EVALUATION OF THE EFFECT OF L-CARNITINE ON LEARNING AND MEMORY IMPAIRMENT INDUCED BY SLEEP DEPRIVATION IN RATS

Sleep deprivation is occurred when
not having enough sleep partially or totally. And its impact on memory
impairment has been related to oxidative stress damage. L-carnitine is a
naturally occurring compound, synthesized endogenously in all mammalian species,
and known to possess antioxidant properties which have a role in brain
neuropathology. In this study, the effect of L-carnitine on learning and memory
impairment in sleep deprived rats was investigated. Sleep deprivation was
induced in animals using modified multiple platform model. L-carnitine was
administered (300mg/kg/day) intraperitoneally for six weeks. Radial arm water
maze (RAWM) was used to assess spatial learning and memory. Additionally, brain
derived neurotrophic factor (BDNF) level, thiobarbituric acid reactive
substance (TBARS) level, and levels of antioxidant defense biomarkers
glutathione reduced (GSH), glutathione oxidized (GSSG), GSH/GSSG ratio,
glutathione peroxidase (GPx), catalase, and superoxide dismutase (SOD) were
assessed in the hippocampus. The results showed that chronic sleep deprivation
impaired both short- and long-term memory, while L-carnitine treatment protect
against this effect in sleep deprived rats. Furthermore, L-carnitine normalized
chronic sleep deprivation induced reduction in the hippocampus activity of
Catalase, GPx, and SOD (p<0.05). Also the ratio of GSH/GSSG in chronically
sleep deprived rats treated with L-carnitine was enhanced significantly as
compared with only sleep deprived rats, without any significant effect on BDNF
level or TBARS activity. In conclusion, chronic sleep deprivation induces
short- and long-term memory impairment, and treatment with L-carnitine
prevented this impairment probably through normalizing antioxidant mechanisms
in the hippocampus.
