VerbascumsinaiticumBenth (Scrophulariaceae) is widely distributed in Jordan with limited investigation of its chemical composition especially the roots.This study was conducted while the major goal is to isolate and identify new chemical entities from this species that were previously unexplored especially from the roots. We were able to isolate harpagoside and pulverolentoside I. Different spectroscopic and spectrometric techniques including: MS (ESI), HRESIMS, 1D-NMR (1H and 13C) and 2D-NMR (HSQC, HMBC and COSY) have been used in order to identify these molecules. Moreover, the volatile oil was analyzed from roots and leaves and the constituents revealed that; 1, 2-Z-propenyl phenol (13.37%), n-undecane(21.53%) and nonanoic acid(21.51%), were the major components of volatile oil extracted from the roots and leaves (first and second trials), respectively. The ethanolic extract was tested for its general cytotoxicity using BSLT, LC50 was found 250 ppm which is considered an active value. Alcoholic extracts (ethanol and methanol) and essential oil both extracted from the roots were evaluated for antibacterial activity against four bacterial strains. The results showed that neither alcoholic extracts nor essential oil have antibacterial activities against any of bacterial strains used.
