sleep deprivation is one of common problems in our society. Studies indicated a direct relationship between sleep deprivation and impairment of learning and memory by increasing oxidative stress in hippocampus caused by accumulation of reactive oxygen species during the wake cycle. Selenium is one of the body's anti-oxidants, which reduces the formation of free radicals and play a major role in glutathione peroxidase enzyme. The East Mediterranean Sage (Salvia triloba) has been reported in old Arabic literature to improve the mental power. Leaves extracts have been shown to process anti-inflammatory, and anti-oxidant activities. According to anti-oxidant activities of these two materials, our studies hypothesize that chronic administration of selenium and sage can antagonize negative effects of sleep deprivation on learning and memory. This hypothesis was evaluated by using multiple platform model to induce sleep deprivation in rats. Selenium (100 µg/100g) and Sage (375mg/Kg) were administered to rats via oral gavage for 6 weeks. Using radial arm water maze behavioral test for short and long term memory were done. BDNF levels and antioxidant markers (GSH, GSSG, GSH/GSSG ratio, GPx, SOD, and catalase) were assessed after hippocampus dissection at the end of the experiment. Also Se levels were determined in many organs (liver, lung, heart, spleen, kidney, and brain). The results of this study indicate that short and long term memory were impaired by chronic sleep deprivation (P<0.05), while Se and Sage administration prevented this effect. Moreover, Se and Sage normalized antioxidants activity which was reduced by CSD such as: Catalase (P<0.05), and SOD (P<0.05), and significantly enhanced the ratio of GSH/GSSG in sleep deprived rats (P<0.05), without any significant effect on BDNF (P>0.05), GSH (P>0.05), or TBARS activity (P>0.05). Se was concentrated in the kidney as it is an exaction organ for it. The highest Se concentration was in Se group in the kidney, where the lowest level was in the control group in the brain. Se distribution was in the following order: Kidney> liver> heart> spleen> lung> brain. P values were calculated indicating significant difference in selenium level in the brain and the kidney. In conclusion, chronic sleep deprivation induced memory impairment, and chronic treatment with Sage and Se prevented this impairment by normalizing antioxidant enzymes activities in the hippocampus.
