Grapes and its products contain high amounts of
polyphenolic compounds, which are known for their profound antioxidant
activities. One of the most potent antioxidants known to occur mainly in the
skin and the seed of grapes is resveratrol, which is a trans-3,5,4´-trihydroxy
stilbene with an antioxidant, anticancer, and phytoestrogenic activities.
A simple, rapid,
selective, and sensitive LC-MS/MS method was developed and validated according
to ICH guideline for the quantification of resveratrol in seed and skin
extracts of grapes. A total of 31 samples were collected from different
locations in Jordan. Chromatographic separation was carried out on a phenomenex
C18 column (5 mm; 125 ×
4.6 mm) using a gradient system mobile phase, CH3CN:aqueous 20 mM NH4OAC,
adjusted to pH 4.3 using acetic acid, with constant flow rate of 400 µL/min.
Electrospray ionization in negative ion MRM mode was utilized for mass
detection. Calibration was linear over 7.81-900 µg/L. Intra- and inter-day
accuracy and precision were within acceptable limits over the range. The
detection limit was calculated as 1.48 µg/L and quantitation limit was
4.49 µg/L to quantify resveratrol in skin and seed extracts of grapes.
Resveratrol content was ranged in seed and skin extracts from 0.011 to 11.6 and
from 0.039 to 3.95, respectively.Baladi cultivar from Karakand Cross Blade from
Maan showed the highest resveratrol content in the skin and seed extracts with 11.6
and 3.95 µg/g dry weight, respectively.
The antioxidant
activities and total phenolic contents ofseed, skin, and whole berry extracts
of the different collected grape cultivars were evaluated using the DPPH radical scavenging assay and Folin–Ciocalteu
colorimetric method, respectively.Seed extracts showed the highest antioxidant
activity and total phenolic content. The total antioxidant capacity of seed,
skin, and whole berry extracts of the investigated cultivars ranged from 49.75 to 68.80, from 4.48 to 49.27, and from 8.23 to
57.43%DPPH inhibition/100 mg dry weight, respectively. While the total phenolic
content ranged from 31.50 to 152.20, from 3.87 to 43.96, and from 3.32 to 33.74
mg GAE/g dry weight, respectively.
Total soluble solidswere
measured for all cultivar juices to investigate the maturity and ripeness of
grapes. Most grape samples analyzed were of good to excellent quality as
reflected by their ⁰Brix values. Brix degree ranged from 5.1 to 22.2%.
