Etazolate is a pyrazolopyridine compound, which exhibits a spectrum of pharmacological mechanisms. Etazolate has a GABA modulating properties and this can illustrate its anxiolytic effect. As a phosphodiesterase 4 (PDE 4) inhibitor and a GABA modulator, etazolate showed positive results in the treatment of depression, traumatic brain injury (TBI) and Alzheimer disease (AD) in animal models and clinical trials. This new drug is still under investigation, thus we found it useful to study the genotoxic effects of etazolate on human cultured lymphocytes. In this study, we evaluated the effects of etazolate on the DNA by measuring the frequency of sister chromatid exchange (SCE) and chromosomal aberrations (CAs) in human cultured lymphocytes. Results of this study showed that all etazolate concentrations which were added to human cultured lymphocytes (0.01 µM, 1 µM, 10 µM, 50 µM) increased the level of SCE in a dose dependent manner. On the other hand, etazolate did not induce CAs in human cultured lymphocytes at all concentrations. Additionally, the levels of 8 hydroxydeoxyguanosine (8-OHdG) did not significantly changed in comparison with the control; this means that etazolate is not associated with oxidative DNA damage. These findings indicate that etazolate may be genotoxic to human lymphocytes and this still needs further confirmatory tests.
